二氫葉酸還原酶分子式:CAS號:性質:是催化葉酸還原成四氫葉酸(H4FA)的酶。H4FA形成過程分兩步進行,由同一個酶催化,H4FA作為一碳單位載體,為嘌呤核苷酸從頭合成和胸嘧啶核苷酸合成等提供一碳單位。氨基蝶呤和氨甲蝶呤的結構類似葉酸,與二氫葉酸還原酶結合的平衡常數為10-10mol/L,比葉酸與該酶的結合能力強1000倍,故是一種極強的競爭性抑制劑。腫瘤細胞經用氨甲蝶呤處理,增殖停止,最終細胞死亡,正是因為核苷酸合成受阻,進而阻斷核酸合成的結果。
