煙草花葉病毒分子式:CAS號:性質:是人類首次提純而得到病毒結晶,它是一種機械性傳播的單鏈:RNA病毒,也是寄生范圍廣泛的植物病原病毒。呈管狀病毒粒子(分子量400 000),約為15×300nm,沉降系數約200。由RNA(占總量5%)和蛋白質(占總量95%)組成,RNA由約6300個核苷酸組成螺旋狀結構(直徑4nm,螺距約2.5nm),大約2100個完全相同的蛋白質衣殼亞基與RNA內外連接形成螺旋結構。蛋白質亞基的裝配從RNA的5端開始。每個蛋白亞基(分子量17 500)由一條多肽鏈組成,其中天門冬氨酸、谷氨酸、絲氨酸和蘇氨酸殘基比例較大。由TMV分離的組分(RNA和蛋白質)體外仍然可以重新構組。用硫酸銨沉淀感染TMV的植物組織,然后進行差速離心可制備純化的TMV。在干葉組織中,TMV的感染力可維持1年以上。此病毒的熱鈍化溫度約為95℃,超聲波、pH值低于3或高于8和化學試劑(如甲醛、碘或尿素溶液)也可使其鈍化。產生局部病斑的寄生有心葉煙和四季豆等。
