技術領域
本發明涉及一種生物技術領域的基因,具體是一種激發水稻產生抗病反應的 無毒基因3KTxa5。
背景技術
水稻白葉枯病由水稻黃單胞菌(Za/7t/ o/z cwas o/y^je pv. o/yzae)引起, 是我國水稻生產上重要的三大病害之一,常造成水稻減產5%-25%,嚴重時會導 致水稻絕收,嚴重制約著我國水稻的安全生產。白葉枯病菌與水稻互作符合"基 因-對-基因"關系,即在病原菌中存在一個無毒基因,則在水稻中存在一個匹配 的抗病基因。白葉枯病菌中的無毒基因可以激發水稻中的抗病基因活性,從而使 水稻產生抗病性。目前為止,在水稻白葉枯病菌中已經鑒別和明確無毒基因功能 的基因有a^fe7, a^fe"和a^fei7,它們分別對應水稻中的抗病基因是Ja7, Xa^ 和Xai^,但只有Ja^7抗病基因被鑒別出來。aKrJa7、 aKrJa70和aKrJai7 都屬于水稻白葉枯病菌中的aw^s3/尸t/^家族。白葉枯病菌中的無毒基因具有植 物抗病育種價值,而且具有分離和鑒別水稻抗病基因的功能,發掘可使水稻產生 抗病性的水稻白葉枯病菌中無毒基因具有重要的科學價值和實踐價值。
經對現有技術的文獻檢索發現,未發現與水稻隱性抗病基因,a5對應的無毒 基因ai^ra5"有關的報道。

發明內容
本發明的目的在于提供一種激發水稻產生抗病反應的無毒基因aw;ra5。本 發明的無毒基因aKrjra5可以激發含有xa5基因的水稻產生對白葉枯病的抗病性。
本發明是通過以下技術方案實現的,本發明涉及的激發水稻產生抗病反應的
無毒基因3Kr;ra5具體是
本發明涉及的無毒基因sktxs5,其序列為SEQ ID NO. 1,序列長度為3717bp,
基因上的酶切位點與aK/^^/Z^^家族基因相同,102bp重復單元在基因內重復
數為19.5。
所述無毒基因SFr;ra5編碼1238個氨基酸,氨基酸序列為SEQ ID NO. 2,由102bp編碼的34個氨基酸重復單元在該蛋白中重復19. 5次。
所述無毒基因sw^a^在含有義s5基因的水稻上可激發產生對白葉枯病的抗病性。
本發明將水稻白葉枯病菌JXOIII菌株的基因組DNA通過fcoRI部分酶切, 構建在pUFR034載體上獲得了 JXOIII基因文庫,以aKr5sJ/Z^A4家族基因的保 守AcoRI-feMH片段為探針,Southern雜交篩選JXOIII基因文庫,獲得了含有 aw^^/尸f力力家族基因的陽性克隆。將陽性克隆逐一導入到水稻白葉枯病菌 PX099A菌株中,在水稻近等基因系成株期水稻上逐一剪葉接種相應的接合子,15 天后調查白葉枯病發病情況。調查發現,p46克隆能夠使PX099A菌株在含有;ra5 抗病基因的IRBB5水稻上由親和性互作轉變為非親和性互作,確定p46克隆中含 有aKTJ^5基因。經過亞克隆、BamHI酶切分析和Southern雜交驗證,獲得了 p4B 亞克隆。將p4B克隆導入PX099A菌株,剪葉接種IRBB5水稻,發現該亞克隆仍 能夠使PX099A菌株在含有;^5抗病基因的水稻品種表現為非親和互作關系,確 認p4B亞克隆中含有一種能夠激發水稻產生抗病反應且對應x35基因的無毒基因 s^ra5。序列測定分析后發現,p4B中含有aKTX35基因,符合aKr^^/尸t力//家 族基因的基本特征。
與現有技術相比,本發明具有如下有益效果將本發明獲得的a^ra5基因 導入水稻白葉枯病菌PX099A菌株中,可使含^^抗病基因的水稻產生抗病性反 應,白葉枯病菌不能侵染含有xa5抗病基因的水稻,從而有效防治白葉枯病菌對 水稻的危害。
本發明中所涉及的菌株JXOIII和PX099A己在文獻《Wu Xiaomin, Li Yurong,
Zou Lifang, and Chen Gongyou. 2007. Gene_for-gene relationships between
rice and diverse avrBs3/pthA avirulence genes in Xanthomonas oryzae pv.
oryzae. Plant Pathology, 56(1): 26—34》中公開。

具體實施方式
下面結合具體實施例,進一步闡述本發明。這些實施例僅用于說明本發明而 不用于限制本發明的范圍。下列實施例中未注明具體條件的實驗方法,通常按照 常規條件,例如Sambrook等分子克隆實驗室手冊(New York: Cold Spring Harbor Laboratory Press, 1989)中所述的條件,或按照制造廠商所建議的條件。
實施例
歩驟一,水稻白葉枯病菌awvra5基因的克隆
水稻白葉枯病菌JXOIII菌株在含有xs5抗病基因的水稻IRBB5上表現為非 親和性互作。本研究以JXOIII為目標菌株,提取其基因組DNA,通過化oRI部 分酶切后構建于載體pUFR034載體上,獲得了 JXOIII基因文庫。圖1中,以 aw^sJ/,fA4家族基因保守的NLS區域DNA片段為探針,Southern雜交篩選 JXOIII基因文庫,獲得了含有家族基因的陽性克隆。圖1中,B 代表^3/in酶切位點,P代表尸stl酶切位點,S代表5^M酶切位點;縱隔表示 102bp重復單元,該單元在arrzs^基因中重復19. 5次;LZ代表亮氨酸拉鏈結構 域;NLS代表核定為信號,1,2,3表示NLS有3個;AD代表酸性轉錄激活域。
將陽性克隆逐一導入到水稻白葉枯病菌PX099A菌株中,在水稻近等基因系成株 期水稻上逐一剪葉接種相應的接合子,15天后調査白葉枯病發病情況。調查發 現,p46克隆能夠使PX099A菌株在含有^5抗病基因的水稻品種IRBB5上由親和 性互作轉變為非親和性互作,如圖4所示,確定p46克隆中含有對應m5的無毒 基因。
步驟二,水稻白葉枯病菌aw7a5基因的分離
對P46克隆中的1B-5B的5個Sa/zHI條帶進行回收,分別連接到經汲mHI 酶切的pUAV45K(B)載體上,獲得了相應的克隆plB、 p2B、 p3B、 p4B和p5B,如 圖2A所示。如圖2B所示,酶切和Sorthern雜交結果顯示,plB和p4B 克隆中含有arrfoJ/尸^4家族的基因。圖2中,圖2A為樣品1-8經皮』1酶切 后的瓊脂糖電泳圖,圖2B為對應樣品的Southern雜交圖。M為DNA Marker, 泳道1為p46,泳道2為plB,泳道3為p2B,泳道4為p3B,泳道5為p4B,泳 道6為p5B,泳道7為pUFR034,泳道8為pUAV45K。
步驟三,水稻白葉枯病菌s7r;^5基因的功能鑒別
為了證實上述2個克隆對含有義s5抗病基因的水稻具有激發抗病性的功能, 本研究將PlB和p4B克隆分別導入PX099A菌株中,獲得了接合子PX099A(plB)和
5PX099A(p4B)。用無針頭注射器將上述接合子注射接種到苗齡為15天的含有zs5 的IRBB5水稻和含有la5的IR24水稻上。褐色過敏反應為抗病反應,水浸癥狀 為感病反應。
圖3A為a^^^基因在水稻上產生水浸癥狀能力的測定,接種的菌株分別為 1為菌株PX099A, 2為菌株JXOIII, 3為菌株PX099A(p46) , 4為菌株PX099A(plB), 5為菌株PX099A(p4B), 6為菌株PX099A(pUFR034)。 IR24為含有Zs5"基因的水稻 品種,IRBB5為含有iaS基因的水稻品種。
如圖3A所示,3天后發現,與JXOIII菌株和PX099A(P46)—樣,PX099'(p4B) 可在IRBB5的水稻上形成褐色過敏反應,而PX099A(plB)與同PX099A—樣僅能形 成水浸(water soaking)癥狀。在含有顯性抗病基因化5的水稻IR24上,無論 是JX0III菌株還是以PX099A為遺傳背景的接合子,則表現為水浸癥狀,如圖3A 所示,表明p4B中的無毒基因不是對應A5基因而是對應犯5基因。
圖3B為水稻白葉枯病菌在水稻IRBB5上生長能力的測定,圖3B中,a為菌 株PX099A(pUFR034), b為菌株JXOIII, c為菌株PX099A(p46) , d為菌株 PX099A(plB), e為菌株PX099A(p4B)。如圖3B所示,和JXOIII菌株一樣,接合 子PX099A(p46)和PX099A(p4B)在水稻組織中的生長能力顯著下降,這表明含有 ^^基因的水稻因被激活了抗病性從而阻止了病菌的生長,進而標明對應罰5基 因的無毒基因為aKrxa^基因。
為了進一步明確aKrxa5在成株期水稻上的無毒基因功能,本研究將上述菌
株和接合子通過剪葉接種方式,分別接種在含有^^隱性基因和含有^J^顯性抗
病基因的水稻IRBB5和IR24上。圖4A為含有xs5隱性基因的水稻品種IRBB5;
圖4B為含有Zs5顯性抗病基因的水稻品種IR24。相應的水稻白葉枯病菌剪葉接
種成株期水稻,根據病斑長度確定aw罰5基因激發水稻產生抗病性的能力,相
應的接種菌株分別為1為菌株PX099A, 2為菌株JXOIII, 3為菌株PX099A(p46),
4為菌株PX099A(plB), 5為菌株PX099A(p4B), 6為菌株PX099A(pUFR034) , 7為
菌株PX099A(aKrZa;)。
結果發現,如圖4A所示,JX0in菌株、PX099A(p46)和PX099A(p4B)均能在
IRBB5上表現為非親和性互作,病斑長度不超過0. 5cra,而在IR24上表現為親和
性互作,病斑長度超過15cm,如圖4B所示。這表明,p4B中的無毒基因激發了
水稻IRBB5中抗病基因化5的活性,從而導致IRBB5水稻產生了抗病性。本發明獲得的aKTX35基因導入水稻白葉枯病菌PX099A菌株中,可使含J^5 抗病基因的水稻產生抗病性反應,使得白葉枯病菌不能侵染含有^^抗病基因的 水稻,從而有效防治白葉枯病菌對水稻的危害。序列表

全文摘要
一種生物技術領域的激發水稻產生抗病反應的無毒基因avrxa5,該基因序列為SEQ ID NO.1,序列長度為3717bp,基因上的酶切位點與avrBs3/PthA家族基因相同,102bp重復單元在基因內重復數為19.5;無毒基因avrxa5編碼1238個氨基酸,氨基酸的序列為SEQ ID NO.2,由102bp編碼的34個氨基酸重復單元在該蛋白中重復19.5次。本發明獲得的avrxa5基因基因可作為水稻抗白葉枯病抗病育種的基因資源,將基因導入水稻白葉枯病菌PXO99<sup>A</sup>菌株后,可使含xa5抗病基因的水稻產生抗病性反應,使白葉枯病菌難以有效侵染水稻。
